These receptors are composed of an extracellular binding domain, a transmembrane lipophilic segment, and an intracellular protein tyrosine kinase domain (deficient in HER3), with a regulatory carboxyl-terminal segment. They become activated upon homo- or heterodimerization, which is promoted either by ligand binding or by high receptor density due to overexpression. The ligands all contain an epidermal growth factor-like domain, and at least six different ligands have been identified for the EGFR.
