  • Gene Introduction:
  • Renal Cell Carcinoma Marker, gp200, is a surface membrane glycoprotein localized to the brush border of the pars convoluta and pars recta segments of the proximal renal tubule and focally on the luminal surface of the Bowman’s capsule adjoining the outgoing proximal tubule. Renal Cell Carcinoma (RCC) Marker expression among other normal tissue types is reported to be restricted to the luminal surface of breast lobules and ducts and epididymal tubular epithelium. Additionally, cytoplasmic expression has been reported in parathyroid parenchymal cells and focal immunoreactivity found in thyroid colloid. No expression has been observed in other normal tissue types tested including adrenal gland, appendix, bladder, bone, cartilage, colon, bile duct, duodenum, endometrium, esophagus, fallopian tube, gall bladder, ileum, jejunum, liver, lung, lymph node, myometrium, ovary, pancreas, prostate, renal pelvis, skin, spleen, stomach, submandibular gland, testis, tonsil, thymus and ureter. Antibodies to RCC Marker are useful in identifying malignant cells from primary and metastatic renal cell carcinomas. RCC Marker expression has been reported in 80-93% of primary renal cell carcinomas and in 67-84% cases of metastatic renal cell carcinoma from sites including adrenal, bladder, bone, brain, breast, liver, lung, lymph node, muscle, pancreas, small bowel, thyroid and trachea. Immunohistochemical detection of RCC Marker may also be utilized for subclassifying renal neoplasms. Several published studies demonstrate that the majority of clear cell [84-92%] and papillary type [93-100%] renal cell carcinomas are RCC Marker positive, however lower positivity rates have also been reported. In contrast, RCC Marker immunoreactivity is much less frequent in chromophobe cell carcinomas [0-45%] and only rare focal reactivity has been observed in renal oncocytoma [0-14%]. Expression of RCC Marker in tumors other than renal cell carcinomas has been reported in 10/22 [45%] intraductal and infiltrating ductal carcinomas of the breast and in 7/8 [88%] In situ and infiltrating lobular breast carcinomas.
