It contains three extensive hydrophobic regions which pass through the cell membrane four times. In addition, the long carboxyl and amino ends of the molecule are located intracellularly with only a minor portion of the molecule exposed outside the cell. The CD20 molecule is involved in regulation of B cell activation, proliferation and differentiation. The CD20 antigen is expressed early during pre-B cell development, presumably just before the expression of cytoplasmic μ-chains, and it persists until plasma cell differentiation. Weak expression of CD20 may be seen in a subpopulation of T cells but the antigen is not expressed in normal myeloid, erythroid, monocytic or mesenchymal cells.
