  • Antibodies against CD66 recognize the carcinoembryonic antigen (CEA) family of proteins and may be further characterized by reactivity with one or more family members indicated by a low letter after CD66: e.g. CD66a (BGP), CD66b (CGM6, CD67), CD66c (NCA), CD66d (CGM1), CD66e (CEA), CD66f (pregnancy-specific glycoprotein (PSG)). The human CEA gene family consists of 22 closely related genes belonging to the Ig superfamily that is located on chromosome 19. The gene family can be divided into two main subgroups, with 9 belonging to the CEA subgroup and 13 to the PSG subgroup. The molecular weight of the different CD66 types varies between 35 kDa and 200 kDa: CD66a (140-180 kDa), CD66b (95-100 kDa), CD66c (90 kDa), CD66d (35 kDa), CD66e (180-200 kDa), and CD66f (found in serum 54-72 kDa). In peripheral blood samples CD66a, CD66b, CD66c and CD66d are only expressed on granulocytes. CD66a, CD66c and CD66e are expressed on epithelial cells as well. Anti-CD66a, CD66abce, CD66ace, CD66acd, CD66acde, and CD66c show binding to lymphoid cell lines of B and T cell origin, childhood ALLs, CML, and AML. Anti-CD66abce has in conjunction with other antibodies been found useful for initial evaluation of adult cases of AML and chronic myeloproliferative disorders, and for detecting minimal residual disease in ALL. The function of the different CD66 molecules is unclear but they are capable of homophilic and heterophilic adhesion, and of activating neutrophils.
