Cloning of the CD22 gene has revealed that CD22 is a member of the IgG superfamily and homologous to several other proteins, including myelin basic protein and members of the carcinoembryonic antigen (CEA) family. The structural features of CD22 suggest that CD22 would mediate intercellular adhesion, however, further work isneeded to understand the in vivo function of CD22 and to identify physiologically significant ligands. CD22 expression is restricted to normal and neoplastic B cells and is absent from other haemopoietic cell types. CD22 is first expressed in pro-B and pre-B cells, largely as a cytoplasmic protein, and only later at the mature B-cell stage is it found on the cell surface. The antigen is lost during the terminal stages of differentiation prior to the plasma cell stage.
