This assay uses the sandwich enzyme immunoassay technique in combination with the enzyme-substrate chromogenic reaction to quantify the analyte in the sample. The color develops positively to the amount of PL in samples. The color intensity is measured at 450 nm via a microplate reader. PL is usually secreted by the pancreatic acinar cells and transferred to the duodenum to participate in the hydrolysis and digestion of fat, cholesterol esters, and fat-soluble vitamins. It is critical for the digestion and absorption of dietary fats and plays an important role in the regulation of gastric emptying and pancreatic and biliary secretion after ingestion of fat in humans. Plasma levels of pancreatic lipase are routinely measured in the diagnosis of pancreatic dysfunction.
