This assay uses the sandwich enzyme immunoassay technique in combination with the enzyme-substrate chromogenic reaction to quantify the analyte in the sample. The color develops positively to the amount of FETUB in samples. The color intensity is measured at 450 nm via a microplate reader. FETUB is a highly selective and potent inhibitor of metallo­peptidases (MPs) of the astacin family. It is a liver-derived plasma protein with serum concentrations of about 0.01 g/l in humans. FETUB plays an essential role in female fertility. By blocking ovastacin, fetuin-B counteracts premature zona pellucida (ZP) hardening before fertilization and therefore maintains female fertility by keeping the ZP penetrable to sperm. Studies have found that FETUB is elevated in type 2 diabetes patients and FETUB knockdown improves glucose metabolism in obese mice.
