It is produced as a 329 AA pre-cursor, which is cleaved to its active form with a length of 215 AA (MW 23,495) by osteoclasts, when they are resorbing bone. Cathepsin K degrades the collagen matrix of the bone after removal of its mineral components. It is actually the most abundantly synthesised protein of the active, resorbing osteoclast and therefore the most specific marker of resorption activity. Cathepsin K plays a key role in tissue destruction, remodelling and cartilage breakdown of the bone.
