  • Beta-2 Glycoprotein 1 (β2GP1), commonly known as Apolipoprotein H (ApoH), is a highly glycosylated protein (MW = 54,200) that is expressed in the liver and then secreted into the plasma. The protein binds to several types of lipoproteins, the most notable of which is cardiolipin. Upon binding to cardiolipin, the structure of the protein complex changes to expose a stretch of amino acids that readily binds phospholipids and other anionic substances, such as heparin, and dextran sulfate.β2GP1 appears to play multiple roles in lipid metabolism, though the precise mechanisms are not clearly understood as yet. Various genotypes show different correlations (or lack of correlations) between β2GP1 levels and either plasma lipid levels or rates of lipid clearance. However, there is considerable evidence that levels of β2GP1 in plasma, or the levels of β2GP1 antibodies, are associated with certain diseases such as liver cirrhosis, antiphospholipid syndrome (APS, thrombosis), and even some form of diabetes. For example, the reduction in plasma levels of β2GP1 correlate with severity of chronic liver disease. There is also evidence that β2GP1 is involved in the regulation of the coagulation system and the generation of antiphospholipid autoantibodies. The multiple roles of β2GP1 in various physiologic pathways clearly underscore the importance of accurate and reproducible assay methods for determining the concentration of β2GP1 in various biological fluids.
