A general feature of systemic rheumatic diseases is the presence of circulating antibodies to a variety of cellular antigens. The detection of these autoantibodies plays a key role in the differential diagnosis of autoimmune diseases such as systemic lupus erythematosus (SLE), rheumatoid arthritis (RA) and mixed connective tissue disease (MCTD). Antibodies to dsDNA are highly specific for SLE and are detected at a high frequency in untreated patients with active disease. The presence of antibodies to dsDNA in SLE has been included as one of the criteria for disease classification by the American Rheumatism Association. Furthermore, several investigators have reported a correlation between disease activity and fluctuations in antibody levels.
