  • Gene Introduction:
  • There are five classes of mammalian immunoglobulins (Ig): IgA, IgD, IgE, IgG, and IgM. In cows, the Ig classes that have been identified are IgM, IgA, IgG1, IgG2 and IgE. No evidence of bovine IgD has been found (Naessens, 1997). Bovine IgA occurs as a single class, whereas in humans, there are two subclasses: IgA1 and IgA2 (Butler, 1983). IgA is the major immunoglobulin found in secretory fluids such as saliva, tears, nasal secretions and intestinal secretions. In rodents and primates, the chief immunoglobulin in colostrum is secretory IgA; however in most domestic animals, including bovine, IgG predominates while IgA is a minor component (Tizard, 1982). IgA is also the most abundant immuoglobulin in the respiratory and intestinal mucosa where it serves as a first line of defense against bacterial and viral invasion from the external environment. Secretory IgA is mainly formed as dimers and higher multimers that are linked via a Jchain linked near the Fc-termini. Secretory IgA also contains a 70 kDa polypeptide associated with the alpha heavy chain, the secretory component (SC), which has been shown to protect IgA molecules from protease activity (Tizard, 1982). IgA is also present in the serum, but in a monomeric form where it functions mainly in the inflammatory response via its interactions with receptors specific for the IgA Fc region (FcalphaR) on effector immune cells (Snoeck, Peters, & Cox, 2006). Reported levels of IgA in cattle serum range from 0.06 – 1.0 mg/ml; in colostrum, levels range from 1.8-14.5 mg/ml; in nasal secretions, levels range from 1.76-3.9; in saliva, levels range from 0.17-0.56 mg/ml; and in tears, levels range from 0.006-.39 mg/ml (Butler, 1983).
