Gene Introduction: Prostatic acid phosphatase (PAP) belongs to the group of acid phosphatase isoenzymes (EC 3.1.3.2). It is found in large amounts in the prostate, and the concentration in seminal fluid is approximately 1 g/L. PAP is synthesized in the secretory epithelial cells in the normal prostate gland. The physiological function of PAP is not known. Chemically it hydrolyzes a wide variety of phosphomonoesters under acidic conditions and is inhibited by L-tartrate. These features make PAP belong to the group of tartrate-inhibitable acid phosphatases, which also includes lysosomal acid phosphatase. In highly differentiated tumours of prostatic origin, an abundance of cells are labelled by antibodies to PAP, whereas PAP-immunoreactive cells are less abundant in moderately (grade I) and poorly differentiated (grade II) carcinomas. The tumour cells of these grade I and II carcinomas also show a greater staining variability. Immunocytochemical data suggests that detection of PAP is a relatively sensitive and specific test for the prostatic origin of an otherwise unclassifiable primary or metastatic neoplasm.
