  • There are five classes of mammalian immunoglobulins (Ig): IgA, IgD, IgE, IgG, and IgM. In cows, the Ig classes that have been identified are IgM, IgA, IgG1, IgG2 and IgE. No evidence of bovine IgD has been found (Naessens, 1997). IgM is the second most abundant Ig in the serum of most animals. The general immunoglobulin structure is described as two heavy and two light chain (H2L2) polypeptides linked together by disulfide bonds. The IgM heavy chain is a mu-type chain and the light chains are of the kappa and lambda type. IgM follows the general H2L2 structure but can exist as a secreted pentamer or as a membrane bound monomer expressed on the surface of mature virgin B cells. The pentameric form consists of 5 covalently linked H2L2 subunits that polymerize end-to-end via inter-mu-chain disulfide bridges and the incorporation of a Jchain (Tizard, 1982). IgM is the first class of antibody to appear in response to a foreign antigen. Since IgM is a pentamer, it is polyreactive and an avid binder. IgM is also a powerful activator of the complement system (Kimball, 1990). IgM is present in bovine secretions and like IgA can complex with the secretory component (SC). Levels of IgM in bovine colostrum are often reported to be higher than IgA; however IgA predominates in other secretory fluids such as nasal secretions, tears, and saliva (Butler, 1983). Concentration of IgM in serum of cattle is reported to be in the range of 0.6 - 4.3 mg/ml (Butler, 1983).
