Gene Introduction: The human VEGFA gene, located on chromosome 6p21.3, contains 8 exons separated by 7 introns. Alternative exon splicing results in generation of 6 different isoforms, VEGF-121, VEGF-145, VEGF-165, VEGF-183, VEGF-189, and VEGF-206, which are named according to the number of amino acids in the peptide chain. Native VEGF protein is a heparin-binding homodimeric glycoprotein of 45 kDa. VEGF-121 is a weakly acidic polypeptide that fails to bind to heparin, while VEGF-165 and VEGF-189 are more basic and bind heparin with increasingly greater affinities. Of these 3 isoforms, VEGF-165 is the predominant molecular form produced in normal cells and tissues. The other isoforms, VEGF-121 and VEGF-189, are detected in the majority of cells and tissues that express the VEGF gene.
